The 2009/2010 season of Sparta Rotterdam was the 54th year in Dutch professional football for the club from Rotterdam. The club played in the Eredivisie and participated in the tournament for the KNVB Cup.
Sparta finished sixteenth in the final ranking and was therefore condemned to the play-offs. In the decisive match against fellow city-dweller SBV Excelsior, Guyon Fernandez scored the 1-1 in the final seconds, which meant that Sparta was relegated again after five years in the Eredivisie. Shortly before that, Fernandez had missed a penalty, after which Rydell Poepon had given the home team a seemingly comfortable 1-0 lead in the 92nd minute. Two minutes later, however, Fernandez struck and goalkeeper Cor Varkevisser was passed.
In the cup tournament, Sparta lost in the quarter-finals to FC Twente: 0-4. Top scorers of the season were centre forward Rydell Poepon and Erik Falkenburg, both with ten goals.
