The club played in the Eredivisie after Heracles was promoted to the highest division in the spring of 2005. Heracles also took part in the KNVB Cup tournament.
The club was led by Peter Bosz, the successor of Gertjan Verbeek. He had piloted the team to sixth place in the previous season (2009/2010). Under Bosz's leadership, Heracles Almelo finished in eighth place in the final ranking. In the subsequent play-offs for participation in the qualifying round for the UEFA Europa League, the club then lost over two matches to FC Groningen.
In the cup tournament, the club suffered by losing in the fourth round to the amateurs of Achilles '29: 5-3. Heracles started at full strength in Groesbeek and after half an hour prepared for an easy evening. Thanks to hits from Darl Douglas and Everton, the formation of trainer Bosz led comfortably 2-0. But the Tukkers misjudged the resilience of the top class player from Gelderland. Achilles '29 came back on the stroke of half-time via a penalty from Tim Verhoeven and Ivo Ritger completed the catch-up race three minutes into the second half.
Even after Douglas' 3-2 for Heracles, Achilles '29 was not discouraged. Substitute Frank Hol brought the team of coach Eric Meijers alongside again, after which Verhoeven even put the home team on the track of a stunt from a penalty kick in the seventieth minute. Hol determined the final score at 5-3 in the 87th minute. “A bad performance”, said Bosz afterwards. “I can't find the right words to describe what happened here tonight. There is no point in commenting on the two penalties, whether they were justified or not. You should never concede five goals.” Captain Antoine van der Linden announces afterwards that the players will reimburse the costs of the accompanying supporters.
